Undervisnings- periode: Weeks 17-24
Undervisnings- form: Lectures, student presentations, group discussions and computer and laboratory exercises, written reports for evaluation.
Formål: To combine the principles of protein structure at the atomic details with experimental and/or theoretical information in order to obtain an understanding of protein functions and mechanisms at the atomic level.
Indhold: The course is an introduction to the two methods used to study protein structures at atomic resolution, x-ray crystallography and NMR spectroscopy. The focus is on the theoretical background of the two methods, on the experimental techniques used to derive three-dimensional structures of proteins, and on the methods used to analyse, refine and evaluate the resulting structures. The course is a mixture of lectures and group discussions of fundamental topics in the field of structure determination and hands on introduction to protein structure determination software tools.
Målbeskrivelse: To obtain the 12 the student should:
• be acquainted with data analysis of NMR spectra and X-ray diffraction data
• be able to analyze heteronuclear NMR spectra in order to obtain protein backbone NMR assignment, amino acid side chain assignment and assignment of nuclear Overhauser effects in protein NOESY spectra
• Use computer programs for automatic protein backbone NMR signal assignments, for automatic assignments of protein NOESY signals, for structure calculations and refinements of proteins based on NMR measurements and evaluation of protein structures with respect to energy and structural geometry
• Master the basic symmetry properties of crystal lattices
• Use computer programs for the analysis of x-ray crystallization data and protein coordinates in order to analyze the quality of protein structures, calculate electron density maps and dock a ligand into the active site of a protein on the basis of calculated electron density maps and molecular graphics
• Perform the initial steps in the progress of molecular replacement
• Be able to read and discuss recent articles describing protein structure determination by NMR spectroscopy and x-ray diffraction
• Must have been present at all sessions
• Have presented papers for at least two topics in the course
Lærebøger: The course material will be a collection of original papers, extracts from textbooks and notes and will be provided by the individual teachers.
Tilmelding: Via KUnet 15 November - 1 December.
Faglige forudsætninger: A bachelor degree either in Biochemistry, Chemistry or Nano-technology is required. Other applicants may be admitted on the basis of an evaluation of their individual qualifications. Basic knowledge of protein science is a requirement.
Eksamensform: It is a requirement to pass the course to have been present at 80% of the sessions; to have presented papers for at least two topics in the course; to have written and handed in a report for each of the sessions of the course; and to have these reports accepted.
The course is evaluated with internal censorship and with pass/not passed.
Reexam: Students which have fulfilled the 80% presence rule but not have all the reports accepted may pass the course if the reports are accepted no later than two months after the end of the course. Evaluated with internal censorship and with pass/not passed.
Eksamen: Løbende evaluering.
Reeksamen: Genaflevering af opgaver senest 2 mdr. efter at kurset er afsluttet.
